Peningkatan Imunogenisitas Vaksin Inaktif Aeromonas salmonicida Dengan Penambahan Adjuvant Pada Ikan Mas (Cyprinus carpio)
Abstract
Bakteri Aeromonas salmonicida dapat menyebabkan penyakit carp erytrodermatitis pada ikan mas (Cyprinus carpio). Salah satu cara untuk mencegah penyakit ini adalah dengan vaksinasi. Adjuvant diketahui dapat meningkatkan imunogenisitas vaksin. Tujuan dari penelitian ini adalah untuk mengetahui tingkat imunogenesitas vaksin inaktif A. salmonicida dengan penambahan beberapa jenis adjuvant yang berbeda. Vaksin diinaktifasi dengan formalin 1% dan diinkubasikan selama 24 jam pada suhu ruang. Penelitian ini terdiri dari 5 perlakuan yaitu tanpa vaksinasi, vaksinasi tanpa adjuvant , dan vaksinasi dengan penambahan 3 jenis adjuvant yang berbeda (6 ppm Aluminum hidroksida (Al(OH)3), 6 ppm Alumunium Potasium Sulfat (KAl(SO4)2), dan Freud’s Incomplete adjuvant (FIA) (1:1 v/v)). Vaksin diujikan pada 10 ekor ikan mas/perlakuan (berat ±30 gr) dengan cara suntik melalui intra peritoneal (107 sel/ikan). Booster dilakukan dengan metode dan dosis yang sama dengan vaksinasi pertama pada 7 hari setelah vaksinasi (hsv). Pengamatan titer antibodi setiap perlakuan diukur dengan metode mikroaglutinasi pada saat sebelum vaksinasi, 7 (hsv), 14 (hsv) (7 hari setelah booster), dan 30 (hsv). Hasil penelitian menunjukkan bahwa penambahan adjuvant mampu meningkatkan imunogenisitas vaksin inaktif A. salmonicida yang ditunjukkan dengan nilai rata-rata titer antibodi yaitu 25 untuk FIA dan KAl(SO4)2 , 24 untuk Al(OH)3 dan vaksin tanpa adjuvant, sedangkan hasil titer antibodi terendah pada perlakuan tanpa pemberian vaksin maupun adjuvant sebesar 23Downloads
