Imunogenisitas Kombinasi Vaksin Inaktif Whole Cell Aeromonas salmonicida dan Jintan Hitam (Nigella sativa) Pada Ikan Mas (Cyprinus carpio)
Abstract
Furunculosis dan carp erytrodermatitis merupakan penyakit pada ikan yang disebabkan oleh bakteri Aeromonas salmonicida. Vaksin inaktif whole cell A. salmonicida saat ini sedang dikembangkan untuk menanggulangi penyakit tersebut. Penelitian ini bertujuan untuk mengetahui imunogenisitas kombinasi vaksin inaktif whole cell A. salmonicida dengan imunostimulan dari jintan hitam (Nigella sativa) pada ikan mas (Cyprinus carpio). Sebanyak 180 ekor ikan mas dimasukkan kedalam 18 akuarium dengan 6 perlakuan yang berbeda dan diulang sebanyak 3 kali. Perlakuan yang dilakukan terdiri dari kontrol (tanpa vaksin dan jintan hitam), pemberian jintan hitam 5%, pemberian vaksin, vaksin yang ditambah jintan hitam 1%, vaksin yang ditambah jintan hitam 2.5%, vaksin yang ditambah jintam hitam 5%. Vaksin diberikan dengan cara penyuntikan secara intraperitoneal dengan kepadatan bakteri 107sel/ikan dan jintan hitam diberikan melalui pakan sebanyak tiga kali dalam sehari. Pengambilan contoh darah ikan untuk diuji titer antibodi, total leukosit, dan hematokrit ikan dilakukan saat sebelum vaksinasi, 7 hari setelah vaksinasi, dan 7 hari setelah booster. Hasil penelitian menunjukkan kombinasi vaksin inaktif whole cell A. salmonicida yang ditambah jintan hitam 5% mampu meningkatkan titer antibodi ikan hingga 27 dibandingkan kontrol yang hanya meningkat hingga 25 dan tanpa jintan hitam yang hanya meningkat hingga 24, kombinasi ini juga mampu meningkatkan jumlah leukosit pada ikan dengan rendahnya nilai hematokrit dan mampu mengembalikan kondisi tubuh dalam keadaan seimbang dalam waktu 7 hari.
